SIS3 is a cell-permeable inhibitor of Smad3 that selectively inhibits TGF-β1-dependent Smad3phosphorylation and Smad3-mediated cellular signaling. SIS3 reduces TGF-β1-induced type 1 procollagenexpression and myofibroblastdifferentiation in normal dermal fibroblasts as well as scleroderma fibroblasts.
