Dual HPLC purification allows to purify oligonucleotides up to 60 bases long, which could be used for stringent applications such as miRNA, siRNA, antisens and firststrand cDNA synthesis.

Dual HPLC combines 2 HPLC purifications to obtain oligonucleotides with higher purity level (up to 95 %). The first stage consists in purifying the DMT-oligonucleotide on a RP-HPLC system followed by IEX-HPLC purification. This second step efficiently removes oligonucleotides with base modifications and DMT-on failure sequences. This combined method allows to purif oligonucleotides up to 60 bases long, which could be used for stringent applications such as miRNA, siRNA, antisens and firststrand cDNA synthesis.
