Sodium Bicarbonate has been the most commonly used buffer in cell culture media because it is the main in vitro / in vivo physiological buffer. Media with Hank’s Balanced Salt Solution (HBSS) contain a low concentration of Sodium Bicarbonate (350 mg/L) and function best in a closed culture vessel. Media with Earle’s Balanced Salt Solution contain a higher concentration of Sodium Bicarbonate (2200 mg/L) and function best in an open vessel, incubated in an atmosphere supplied with 5% CO2 in air.
