Hematopoiesis is the process by which blood cells are produced. Hematopoietic stem cells (HSC) are generally found in the bone marrow, where they self-renew and begin their journey of differentiating into all the different blood cell types. During differentiation, HSCs progress through various committed progenitor stages before fully differentiating into mature cell types.
While there is some scientific debate around how to characterize hematopoietic stem and progenitor cells (HSPCs) simply by phenotypic markers, most believe the cell types cannot be fully characterized without functional testing of differentiation capacity. Regardless, it is widely accepted that the CD34 marker is indicative of stem and progenitor cells, and thus it is important to assess presence of the CD34 marker when working with HSPCs.
Hematopoietic progenitor cells can be categorized into either myeloid progenitors or lymphoid progenitors. The myeloid progenitor cells are able to give rise to myeloid lineage cells including platelets, eosinophils, basophils, neutrophils, monocytes, and erythrocytes. Lymphoid progenitor cells are capable of differentiating into T-cells and B-cells, which are key players in adaptive immunity.
Hematopoietic stem cells progress down a differentiation pathway to committed progenitors in the bone marrow. At that time, they typically leave the bone marrow and enter the peripheral blood and tissues, where they complete differentiation into the various mature immune cell types.

Normal human bone marrow donors are healthy males and non-pregnant females between the ages of 18 and 45 years old. All bone marrow and peripheral blood donors are assessed for good general health via acceptable vital signs and hematology values and have a negative medical history of heart disease, kidney disease, liver disease, cancer, epilepsy and blood or bleeding disorders. In addition, each donor tests negative for HIV-1, HIV-2, hepatitis B and hepatitis C. Where donor testing is not possible, cell products are tested for the presence of viral nucleic acid from HIV, Hepatitis B Virus, and Hepatitis C Virus.
